Crisantaspase recombinant (JZP-458) is a recombinant form of the enzyme L-asparaginase derived from *Erwinia chrysanthemi*. It is produced using the Pelican Expression Technology platform (formerly Pfenex Expression Technology), which utilizes *Pseudomonas fluorescens* for high-quality protein production. The drug is specifically indicated as a component of a multi-agent chemotherapeutic regimen for the treatment of acute lymphoblastic leukemia (ALL) and lymphoblastic lymphoma (LBL) in adult and pediatric patients who have developed hypersensitivity or silent inactivation to *E. coli*-derived asparaginase products. By depleting circulating levels of L-asparagine—an amino acid that neoplastic cells are unable to synthesize efficiently due to low asparagine synthetase expression—crisantaspase recombinant inhibits protein synthesis and induces apoptosis in leukemic cells. It was developed by Jazz Pharmaceuticals to address manufacturing challenges and supply shortages associated with the naturally derived Erwinia asparaginase (Erwinaze).
