CRISPR G6PD knockout refers to the use of CRISPR/Cas9 gene-editing technology to permanently disrupt the *G6PD* gene, which encodes Glucose-6-Phosphate Dehydrogenase. This enzyme is the rate-limiting step in the oxidative branch of the pentose phosphate pathway (PPP), responsible for regenerating NADPH to maintain cellular redox balance. In the context of oncology research, specifically melanoma, knocking out G6PD has been shown to increase reactive oxygen species (ROS) levels and decrease the metastatic potential of tumor cells by impairing their ability to manage oxidative stress during transit through the bloodstream. This approach is currently a research tool used to validate G6PD as a therapeutic target rather than a clinical-stage drug product.
