Overview

Crizotinib + axitinib is a combination of two small molecule tyrosine kinase inhibitors investigated for the treatment of advanced solid tumors, including metastatic renal cell carcinoma (mRCC). Crizotinib inhibits multiple receptor tyrosine kinases, primarily anaplastic lymphoma kinase (ALK), hepatocyte growth factor receptor (HGFR/c-MET), ROS1, and Recepteur d'Origine Nantais (RON), thereby blocking oncogenic signaling pathways that drive tumor proliferation and survival. Axitinib selectively inhibits vascular endothelial growth factor receptors 1–3 (VEGFR-1, VEGFR-2, VEGFR-3), suppressing angiogenesis and tumor growth. The combination aims to provide greater clinical benefit by targeting both angiogenesis and oncogenic signaling. Clinical studies have shown the combination has a manageable safety profile with evidence of antitumor activity in mRCC[1][3][4][5][6].
