Overview

CRKL siRNA is a research-grade small interfering RNA (siRNA) molecule designed to silence the expression of the CRKL gene, which encodes the CRK-like adaptor protein. CRKL is an adaptor protein involved in various signaling pathways, including those downstream of ALK and BCR-ABL, and is often overexpressed in multiple malignancies. By utilizing the RNA interference (RNAi) pathway, CRKL siRNA induces the degradation of CRKL mRNA, leading to reduced protein levels. In preclinical research, this knockdown has been shown to inhibit cell proliferation, migration, and invasion, and to reverse chemotherapy resistance in models of glioblastoma, non-small cell lung cancer, gastric cancer, and leukemia. It is primarily used as a laboratory tool to validate CRKL as a therapeutic target and is not currently a clinical-stage drug candidate.
