Drug intelligence / Profile preview
CRLF2-FDC is a novel antibody fragment-drug conjugate (FDC) designed to target cytokine receptor-like factor 2 (CRLF2), a cell-surface protein overexpressed in approximately 50% of Philadelphia chromosome-like acute lymphoblastic leukemia (Ph-like ALL) cases. Developed through a collaboration between the Children's Cancer Institute, the Garvan Institute of Medical Research, and Jena University Hospital, the conjugate consists of a single-chain anti-CRLF2 variable monoclonal antibody fragment (scFv-Fc) linked to the cytotoxic anti-microtubule agent maytansinoid 1 (DM1) via a non-cleavable linker. By targeting CRLF2, which plays a critical role in leukemogenesis and cell survival via JAK/STAT signaling, CRLF2-FDC delivers DM1 directly to leukemia cells, inducing apoptosis while minimizing off-target toxicity. In preclinical models of pediatric Ph-like ALL, CRLF2-FDC demonstrated potent anti-leukemic activity both as a single agent and in combination with standard-of-care induction chemotherapy, while also exerting an antagonistic effect on STAT5 signaling.
