CRLX101 + bevacizumab is a combination therapy comprising CRLX101, a nanoparticle-drug conjugate containing camptothecin, and bevacizumab, an anti-VEGF monoclonal antibody. CRLX101 delivers camptothecin using a cyclodextrin-containing polymer nanoparticle allowing sustained intratumoral exposure, which inhibits topoisomerase I and hypoxia-inducible factors (HIF1α, HIF2α)[2][5][6]. Bevacizumab targets and neutralizes vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis[5]. The combination seeks to provide dual inhibition in the hypoxia and angiogenesis pathway—CRLX101 targets HIF1α/HIF2α whose expression drives VEGF production, while bevacizumab blocks VEGF-mediated angiogenesis. This pairing has been investigated in phase I/II clinical trials for metastatic renal cell carcinoma (mRCC) and recurrent ovarian cancer[2][3][6], based on hypothesized synergy in both preclinical and early-phase human studies.
