Overview

CRLX301 is an investigational nanoparticle-drug conjugate (NDC) consisting of **docetaxel covalently conjugated to a cyclodextrin-polyethylene glycol co-polymer** designed to enhance targeted delivery of docetaxel to tumor tissue. It accumulates in tumors and provides controlled, slow release of docetaxel, resulting in greater intratumoral drug concentration and prolonged systemic exposure compared to conventional docetaxel. Preclinical studies showed improved efficacy and reduced toxicity versus standard docetaxel. Clinical studies explored intravenous administration in advanced solid tumors, using both every-three-week and weekly dosing schedules. The mechanism of action is mediated by the release of docetaxel, which stabilizes microtubules and thereby inhibits mitosis and induces cell death.
