Overview

CRN-AntimiR-122 is a conformationally restricted nucleotide (CRN)-substituted antisense oligonucleotide (antagomir) developed by Marina Biotech. It is designed to target and inhibit microRNA-122 (miR-122), a liver-specific microRNA that plays a critical role in liver physiology and is a known host factor for hepatitis C virus (HCV) replication. The CRN modification is a proprietary sugar modification that locks the ribose ring in a specific conformation, which significantly enhances hybridization affinity to the target RNA and increases resistance to nuclease degradation. In preclinical studies, CRN-AntimiR-122 demonstrated a dose-dependent de-repression of liver-specific target genes, including Aldolase A (AldoA), Glycogen Synthase I (GYS1), and Solute Carrier Family 7 member 1 (SLC7A1), while maintaining a favorable safety and tolerability profile.
