Overview
CRN12755 is an investigational small molecule, orally bioavailable, potent functional antagonist of the human thyroid-stimulating hormone receptor (TSHR). It is being developed by Crinetics Pharmaceuticals for the treatment of Graves' disease, including Graves' hyperthyroidism and Graves' orbitopathy (thyroid eye disease). Preclinical studies have shown that CRN12755 reduces TSAb-stimulated thyroid hormone (T4) levels as well as inflammatory markers such as hyaluronic acid and IL-6 in animal models. The agent is in the IND-enabling stage as of June 2025, with no human safety or efficacy data reported to date[1][3][4][5].
