CRO-CD7 CAR-NK cells are an engineered cell therapy derived from human pluripotent stem cells where the CD7 gene is knocked out and a CD7-specific chimeric antigen receptor (CAR) is introduced, followed by overexpression of the CXCR4 chemokine receptor. The knockout of CD7 prevents cellular fratricide during manufacturing and application, as CD7 is expressed on both normal and malignant NK/T cells. CXCR4 overexpression enhances cell homing to bone marrow and tissue persistence. These cells are differentiated into induced NK cells (iNKs) and used as an adoptive cell therapy to target CD7-positive T-cell malignancies, notably T-cell acute lymphoblastic leukemia (T-ALL). The approach enables large-scale generation of fratricide-resistant, highly potent CAR-NK cells with improved anti-tumor efficacy in vivo[1][3].
