Overview

Crocetinic acid is a naturally occurring carotenoid dicarboxylic acid isolated from saffron (*Crocus sativus*). It is the aglycone form of crocin and has been investigated for its potent anti-cancer properties, particularly in pancreatic adenocarcinoma. The compound exerts its effects through several mechanisms, including the inhibition of the Sonic Hedgehog (Shh) signaling pathway (downregulating Shh, Smoothened, Patched-1, and Gli-1), the suppression of the EGFR/Akt signaling axis, and the inhibition of histone deacetylases (HDACs). Research conducted at the University of Kansas Medical Center has shown that crocetinic acid can effectively target pancreatic cancer stem cells (CSCs), as evidenced by the reduction of CSC markers like DCLK-1 and the inhibition of pancosphere formation. In vivo studies have demonstrated significant regression of tumor growth and induction of apoptosis.
