Overview

Crotamiton is a topical scabicidal and antipruritic agent used primarily for the treatment of scabies and relief of pruritus (itching) associated with various skin conditions. It is available as a cream or lotion for topical use only. The drug acts as an anti-parasite by killing the mites that cause scabies, likely through disruption of their nerves and muscles. Its antipruritic effect is attributed to inhibition of sensory ion channels such as transient receptor potential vanilloid 4 (TRPV4), which are expressed in the skin and primary sensory neurons. Additionally, crotamiton relieves itching by producing a cooling effect on evaporation from the skin (counter-irritation). The precise mechanism underlying its scabicidal action remains unknown[1][2][5][8].
