Overview

Crovalimab is a humanized monoclonal antibody that acts as a complement C5 inhibitor. It is used for the treatment of paroxysmal nocturnal hemoglobinuria (PNH) in adults and adolescents. Crovalimab binds with high affinity to the complement protein C5, blocking its cleavage into C5a and C5b, thereby preventing the formation of the membrane attack complex (MAC) and inhibiting terminal complement-mediated intravascular hemolysis. This mechanism reduces red blood cell destruction in PNH patients. Crovalimab employs an anti-C5 sequential monoclonal antibody recycling technology, allowing repeated binding to its target for sustained inhibition. The drug was developed by Chugai Pharmaceutical in collaboration with Roche and has been approved in China, Japan, and the USA for PNH[2][6][7].
