CRPA1A2 is a first-in-class T-cell receptor (TCR)-based bispecific T-cell engager developed by Beijing CorreGene Biotechnology. It is designed to target the cancer-testis antigen melanoma-associated antigen 1 (MAGE-A1) when presented by HLA-A*02:01 molecules on the surface of tumor cells. The therapeutic mechanism involves engaging the T-cell receptor complex (typically via CD3) to redirect and activate T cells against MAGE-A1-positive malignant cells, facilitating immune-mediated tumor cell lysis. MAGE-A1 is frequently expressed in various cancers, including lung squamous cell carcinoma, hepatocellular carcinoma, and esophageal cancer, but has minimal expression in healthy tissues, providing a wide therapeutic window. The program is currently in the IND-enabling phase, with a planned US FDA IND filing in the third quarter of 2025.
