CRPKDA11 is a T-cell receptor (TCR)-based bispecific antibody developed by Corregene Biotechnology, specifically designed to target the KRAS G12D mutation in solid tumors such as colorectal and pancreatic cancers. The molecule functions as a T-cell engager, featuring a high-affinity TCR domain that recognizes the KRAS G12D peptide presented by the HLA-A*11:01 MHC complex on tumor cells, and a CD3-binding domain that recruits and activates T cells. This dual-binding mechanism bypasses the need for natural TCR recognition, allowing for potent and specific T-cell mediated lysis of KRAS G12D-positive tumor cells. The drug targets one of the most common KRAS mutations, which is prevalent in approximately 90% of pancreatic cancers and 40% of colorectal cancers.
