CRS-207 is a live, attenuated, double-deleted Listeria monocytogenes (LADD) engineered to express the tumor-associated antigen mesothelin. It was developed by Aduro Biotech (later associated with Chinook Therapeutics/Novartis) as an immunotherapy to stimulate both innate and adaptive immune responses against tumors that overexpress mesothelin, such as malignant pleural mesothelioma, pancreatic cancer, ovarian cancer, gastric cancer, and lung cancers. The drug works by using genetically modified Listeria bacteria to deliver the mesothelin antigen directly into the body, thereby activating a targeted anti-tumor immune response. Clinical trials have evaluated its use alone or in combination with chemotherapy or checkpoint inhibitors for various solid tumors; however, development of the LADD platform has been discontinued by Aduro.
