Overview

CRS0540 is a novel small molecule antibiotic in preclinical development for the treatment of serious Gram-positive bacterial infections, including those caused by antibiotic-resistant pathogens such as methicillin-resistant Staphylococcus aureus (MRSA), penicillin-resistant Streptococcus pneumoniae (PRSP), vancomycin-resistant Enterococci (VRE), and Listeria monocytogenes. It is a thiadiazole urea compound that acts as a potent inhibitor of PolC, the catalytic subunit of DNA polymerase III essential for DNA replication in Gram-positive bacteria. By targeting this novel bacterial enzyme, CRS0540 circumvents existing mechanisms of resistance and demonstrates broad-spectrum bactericidal activity against clinically significant Gram-positive pathogens. The drug has shown promising efficacy in multiple preclinical models and achieves high intracellular concentrations relative to extracellular levels, which may be advantageous for treating intracellular infections like listeriosis. Developed by Crestone, Inc., CRS0540 is being advanced as both an oral and intravenous agent[1][2][6][7][8].
