Overview

CRT-401 is an investigational in vivo CAR-T cell therapy developed by CREATE Medicines (formerly Myeloid Therapeutics) for the treatment of epithelial tumors. Utilizing the company's proprietary "Multi-immune Cell Programming" platform, the drug consists of mRNA encapsulated in lipid nanoparticles (LNPs). Upon intravenous administration, these mRNA-LNPs are designed to program multiple immune cell types—including T cells, natural killer (NK) cells, and myeloid cells—directly within the patient's body to express a chimeric antigen receptor (CAR) targeting Mucin-1 (MUC1). This approach aims to generate a coordinated, multi-pronged anti-tumor response and overcome the immunosuppressive tumor microenvironment without the need for complex ex vivo cell manufacturing. CRT-401 is currently in Phase 1 clinical development.
