Overview

CRT-402 is an experimental in vivo CAR-T cell therapy being developed by CREATE Medicines (formerly Myeloid Therapeutics) for the purpose of B cell depletion. The therapy utilizes a proprietary "Multi-immune Cell Programming" technology, which employs mRNA encapsulated in lipid nanoparticles (LNPs) to deliver genetic instructions directly into the patient's body. Unlike traditional ex vivo CAR-T therapies that require the extraction and modification of T cells outside the body, CRT-402 is designed to program multiple immune cell types—including T cells, Natural Killer (NK) cells, and myeloid cells—to express a Chimeric Antigen Receptor (CAR) in vivo. This CAR typically targets the CD19 antigen to facilitate the systemic depletion of B cells, making it a potential treatment for B-cell-mediated malignancies and various autoimmune diseases. The drug is currently in Phase 1 clinical development.
