CRx-140

CRx-140 is an investigational oral combination therapy comprising the selective serotonin reuptake inhibitor (SSRI) fluoxetine and the tricyclic antidepressant (TCA) nortriptyline. Developed by CombinatoRx using their proprietary high-throughput screening platform, the product was designed to leverage synergistic interactions between serotonergic and noradrenergic pathways for the treatment of neuropathic pain. The mechanism of action involves the dual inhibition of serotonin and norepinephrine reuptake, which is intended to modulate descending inhibitory pain pathways more effectively than single-agent treatments. It was primarily evaluated for the management of diabetic peripheral neuropathy, but development was halted following Phase 2 clinical trials that failed to demonstrate a statistically significant improvement over nortriptyline monotherapy.
