Overview

CRx-197 is a topical combination drug candidate consisting of the tricyclic antidepressant nortriptyline hydrochloride and the second-generation antihistamine loratadine. Developed by Zalicus (formerly CombinatoRx) using its proprietary combination High Throughput Screening (cHTS) platform, CRx-197 was designed to treat inflammatory skin conditions, specifically plaque psoriasis. The rationale behind the combination is to leverage the synergistic anti-inflammatory effects of its components, which are thought to modulate cytokine production and neurogenic inflammation pathways in the skin. The drug was evaluated in Phase 2a clinical trials as a topical cream, but development was subsequently halted as the company shifted its strategic focus toward ion channel modulators and pain management therapies.
