Cryptophycin-ADC is an antibody-drug conjugate (ADC) being developed by Lantern Pharma in collaboration with Bielefeld University. It utilizes a cryptophycin analog (such as cryptophycin-52 or cryptophycin-55) as a highly potent cytotoxic payload. Cryptophycins are microtubule-disrupting agents that bind to the vinca alkaloid site of tubulin, inhibiting microtubule polymerization and leading to mitotic arrest and apoptosis. This ADC is designed to target a variety of solid tumors, including breast, bladder, colorectal, gastric, pancreatic, and ovarian cancers. Preclinical studies have demonstrated picomolar potency and the ability to overcome resistance to common ADC payloads like monomethyl auristatin E (MMAE). Lantern Pharma is advancing the program toward Investigational New Drug (IND) development for patients with refractory or unresponsive solid tumors.
