Overview

CS-190 is a novel small molecule inhibitor of Leucine-rich repeat kinase 2 (LRRK2) being investigated for the treatment of inflammatory conditions, specifically Crohn's disease. LRRK2 is a genetic hotspot for gain-of-function mutations associated with both Crohn's and Parkinson's disease; these mutations enhance LRRK2 kinase activity, leading to increased inflammatory signaling. CS-190 works by suppressing LRRK2 kinase activity, thereby inhibiting the phosphorylation of downstream targets such as Rab10 and Rab12. In preclinical in vitro studies, CS-190 has demonstrated the ability to suppress TNF-alpha production in human dendritic cells and inhibit NLRC4 inflammasome-mediated production of IL-1beta. The compound was developed through a collaboration involving the Icahn School of Medicine at Mount Sinai and the NIAID.
