Overview

CS 82 is a novel small molecule inhibitor of Leucine-Rich Repeat Kinase 2 (LRRK2) currently under investigation for the treatment of inflammatory bowel disease (IBD) and Crohn's disease. Developed by researchers at the National Institute of Allergy and Infectious Diseases (NIAID) and the Icahn School of Medicine at Mount Sinai, CS 82 works by inhibiting the kinase activity of LRRK2, which prevents the phosphorylation of its downstream targets, Rab10 and Rab12. This inhibition further blocks the activation of the NLRC4 inflammasome and the subsequent release of pro-inflammatory cytokines such as IL-1β and TNFα. Preclinical studies suggest that CS 82 can significantly reduce the severity of intestinal inflammation, positioning it as a potential therapeutic candidate for auto-inflammatory conditions associated with LRRK2 dysregulation.
