**CS-TEMPO-OVA** is an experimental metal-free theranostic cancer nanovaccine developed for melanoma models. The nanoparticle integrates chitosan, the organic nitroxide radical 4-carboxy-TEMPO, and ovalbumin as a model tumor antigen. It promotes dendritic-cell activation, antigen-driven T-cell responses, cytotoxic T-lymphocyte activity, cytokine production, and macrophage M1 polarization, resulting in inhibition of subcutaneous and metastatic B16-OVA melanoma in mice. TEMPO supplies T1 magnetic-resonance-imaging contrast functionality, enabling tumor imaging alongside immunotherapy. In mouse studies, administration by intravenous or intramuscular injection enhanced tumor T1 MRI contrast; combination with anti-PD-1 therapy increased intratumoral cytotoxic T-cell infiltration and strengthened systemic antitumor activity.
