Overview

CS1(dCH2)BBz CAR T cells (also known as MB-104) is an autologous chimeric antigen receptor (CAR) T-cell therapy targeting SLAM family member 7 (SLAMF7, also known as CS-1 or CD319). Developed by City of Hope National Medical Center and previously licensed to Mustang Bio, the therapy utilizes autologous memory-derived T cells transduced with a self-inactivating lentiviral vector to express a CS1-targeting, hinge-optimized, 4-1BB costimulatory CAR, along with a truncated human epidermal growth factor receptor (EGFRt) as a selection marker and safety switch. It is under clinical evaluation for the treatment of relapsed and refractory multiple myeloma.
