Overview

CS1-CAR is a chimeric antigen receptor (CAR) T-cell therapy designed to target the CS1 antigen, also known as SLAMF7 or CD319. CS1 is a cell surface glycoprotein that is highly and nearly universally expressed on malignant plasma cells in multiple myeloma, as well as on normal plasma cells and a subset of natural killer (NK) cells. The therapy involves genetically engineering a patient's T-cells to express a synthetic receptor comprising an extracellular single-chain variable fragment (scFv) specific for CS1, a hinge and transmembrane region, and intracellular signaling domains (typically CD3-zeta and a costimulatory domain such as 4-1BB or CD28). When these engineered T-cells encounter CS1-expressing cells, the CAR binds to the antigen, triggering T-cell activation, expansion, and targeted cytotoxic activity against the tumor cells. CS1-CAR has demonstrated potent anti-myeloma activity in preclinical studies and is being evaluated in clinical trials to address relapsed or refractory multiple myeloma, often as a component of multi-targeted or compound CAR strategies to prevent antigen escape.
