Overview

CS1-CAR NK cells are a type of adoptive cell therapy where natural killer (NK) cells are genetically engineered to express a Chimeric Antigen Receptor (CAR) specifically targeting CS1 (also known as SLAMF7 or CD319). CS1 is a cell-surface glycoprotein that is highly and ubiquitously expressed on multiple myeloma (MM) cells, while its expression is low on normal immune cells and almost undetectable on most healthy tissues. The CAR construct typically includes an extracellular antigen recognition domain (often a single-chain variable fragment, scFv), a transmembrane domain, and intracellular signaling domains (such as CD3ζ and co-stimulatory domains like CD28 or 4-1BB). This engineering redirects the NK cells to specifically recognize and bind to CS1-expressing MM cells, leading to enhanced cytotoxicity and increased production of cytokines like interferon-gamma (IFN-γ). This approach aims to provide a potent anti-tumor response against multiple myeloma, potentially offering a safer alternative to CAR-T cell therapies due to a lower risk of severe side effects like cytokine release syndrome and neurotoxicity.
