Overview

CS1 CAR T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target the CS1 (also known as SLAMF7 or CD319) antigen, a cell surface glycoprotein highly and selectively expressed on malignant plasma cells. Developed primarily by researchers at City of Hope, these second-generation CAR T cells utilize a CS1-specific single-chain variable fragment (scFv) coupled with costimulatory domains such as CD28 or 4-1BB and a CD3ζ signaling chain. The construct often incorporates a truncated human epidermal growth factor receptor (huEGFRt) to facilitate cell tracking and provide a safety mechanism for ablation via cetuximab. CS1 CAR T cells are being evaluated for the treatment of relapsed or refractory multiple myeloma and light chain amyloidosis, with research indicating that their efficacy and persistence can be further enhanced when used in combination with immunomodulatory drugs like lenalidomide or targeted alpha therapy.
