**CS1-dbBiTE** is an experimental, preclinical dual-specific bivalent T-cell-engaging antibody construct developed at City of Hope for **multiple myeloma**. It is produced by click-chemistry conjugation of the anti-CS1 antibody elotuzumab with an anti-human OKT3 antibody at their hinge regions. The construct binds signaling lymphocytic activation molecule family member 7 on malignant plasma cells and CD3 on T cells, bringing T cells into proximity with CS1-expressing tumor cells to induce T-cell activation, degranulation, inflammatory cytokine production, and tumor-cell killing. In preclinical multiple-myeloma xenograft models, ex vivo CS1-dbBiTE-armed human T cells reduced MM.1S tumor burden.
