CS1-targeted CAR-T cells is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy being developed by Zhejiang University in collaboration with Yake Biotechnology for the treatment of relapsed or refractory multiple myeloma. The therapy consists of autologous T cells genetically modified to express a CAR that targets CS1 (also known as SLAMF7 or CD319), a cell surface glycoprotein highly and nearly universally expressed on malignant plasma cells. The CAR construct utilized in this program incorporates an anti-CS1 single-chain variable fragment (scFv) linked to CD28 costimulatory and CD3ζ signaling domains. This therapy is specifically being evaluated in clinical trials for patients who have relapsed after receiving BCMA-targeted CAR-T therapies, as well as those with CS1-positive/BCMA-negative disease, addressing a significant unmet need in advanced myeloma management.
