Overview

CS2009 is a next-generation trispecific antibody developed by CStone Pharmaceuticals for the treatment of solid tumors. It simultaneously targets three clinically validated immune and angiogenesis pathways: programmed cell death protein 1 (PD-1), vascular endothelial growth factor A (VEGFA), and cytotoxic T-lymphocyte–associated antigen 4 (CTLA-4). The molecular design enables preferential targeting of PD-1/CTLA-4 double-positive tumor-infiltrating lymphocytes, reinvigorating exhausted T cells within the tumor microenvironment while neutralizing VEGFA to inhibit angiogenesis. Preclinical studies have demonstrated that CS2009 exhibits superior anti-tumor activity compared to bispecific antibodies or combination therapies targeting these axes individually, with enhanced efficacy in VEGFA-enriched environments and minimized peripheral immune-related toxicity. The drug is positioned as a potential first-in-class or best-in-class immuno-oncology backbone therapy for a broad range of solid tumors, including non-small cell lung cancer, hepatocellular carcinoma, gastric adenocarcinoma, endometrial cancer, ovarian cancer, renal cell carcinoma, and cervical cancer[1][2][3][5][6].
