CS32582 is a novel, orally administered small molecule and highly selective allosteric inhibitor of Tyrosine Kinase 2 (TYK2), independently developed by Chipscreen. It achieves its selectivity by specifically binding to the regulatory pseudokinase JH2 domain of TYK2, without inhibiting other Janus kinase family members (JAK1, JAK2, JAK3) at therapeutic doses. This selectivity is designed to ensure both efficacy and a favorable safety profile. By inhibiting TYK2, CS32582 blocks downstream signaling pathways mediated by cytokines such as interleukin-23 (IL-23), interleukin-12 (IL-12), and type I interferons—key mediators in autoimmune diseases like psoriasis. Preclinical studies have demonstrated significant efficacy in mouse models of psoriasis. The drug is currently being evaluated in Phase 1 clinical trials for the treatment of psoriasis[1][4][5][6].
