CS5007 is a bispecific antibody-drug conjugate (ADC) developed by CStone Pharmaceuticals for oncology applications. It targets both the epidermal growth factor receptor (EGFR) and human epidermal growth factor receptor 3 (HER3), using a bispecific antibody backbone (CS2011). The ADC is constructed with a hydrophilic β-glucuronide linker and delivers the clinically validated topoisomerase I inhibitor, Exatecan, as its cytotoxic payload. This design enables precise targeting of tumor cells expressing EGFR and/or HER3, leading to high-rate internalization and potent antigen-dependent cytotoxicity across various tumor types. Preclinical studies have demonstrated robust antitumor activity, favorable safety profile, superior in vitro stability compared to other ADCs with different linkers, and pharmacokinetics comparable to monoclonal antibodies. CS5007 is positioned as a potential best-in-class candidate for precision oncology in advanced solid tumors[1][3][5].
