The **CSC antigen nanodisc vaccine** is a peptide-based cancer vaccine targeting cancer stem cells (CSCs) using a synthetic high-density lipoprotein (sHDL) nanodisc platform. It is formulated with multiple immunogenic epitopes derived from CSC-associated proteins—Sox2, Nanog (both transcription factors), and aldehyde dehydrogenase (ALDH, specifically ALDH-A1 and ALDH-A3)—together with a Toll-like receptor 9 (TLR-9) agonist (CpG oligonucleotide) used as an adjuvant. The nanodiscs deliver these peptides and CpG to antigen-presenting cells in lymph nodes, inducing robust CD8+ T cell immune responses against CSCs. Preclinical evidence, especially in highly aggressive melanoma models, demonstrates that the vaccine reduces CSC frequency, remodels the tumor microenvironment to be more pro-inflammatory, and significantly delays tumor growth and extends survival. The vaccine is designed as a cost-effective, safe approach to specifically target therapy-resistant tumor-initiating CSCs, potentially for use in multiple cancer types.
