The CSC-DC vaccine is an **experimental cancer vaccine** designed to stimulate the immune system to target cancer stem cells (CSCs) by pulsing autologous dendritic cells (DCs) with lysates of ALDHhigh-enriched cancer stem cells. This approach aims to induce CSC-specific cytotoxic T lymphocyte (CTL) and antibody responses, reduce the population of CSCs within tumors, and block metastasis. Mechanistic studies show the vaccine triggers host immunity specifically against CSCs and may reduce metastatic dissemination by downregulating chemokine receptors (CCR7, CCR10) and their ligands in target organs. Its efficacy has been demonstrated in preclinical syngeneic murine tumor models and is under exploration in combination regimens with immune checkpoint inhibitors (such as anti–PD-L1 antibodies) for enhanced tumor control[1][2][3][6][10].
