Overview

CSF1R-TCB is a T-cell engaging bispecific antibody (TCB) designed to target the colony-stimulating factor 1 receptor (CSF1R) on acute myeloid leukemia (AML) blasts while simultaneously engaging CD3 on T cells. Developed using Roche's CrossMAb® technology in a 2+1 format (two binding sites for CSF1R and one for CD3), it aims to provide a safer alternative to traditional AML targets like CD33 or CD123, which often exhibit on-target-off-tumor toxicity toward hematopoietic stem and progenitor cells (HSPCs). Preclinical studies demonstrate that CSF1R-TCB induces potent T-cell-mediated lysis of AML cell lines and primary blasts with minimal impact on HSPCs and reduced cytokine release compared to CD33-targeted therapies.
