cSNX1.3 is a novel, end-capped cell-penetrating peptide therapeutic developed by researchers at the University of Arizona. It is designed to mimic the BAR domain of Sorting Nexin 1 (SNX1) and competitively inhibit the interaction between SNX1 and the Epidermal Growth Factor Receptor (EGFR). By blocking this interaction, cSNX1.3 prevents the retrograde trafficking and nuclear translocation of EGFR (nEGFR), a non-canonical oncogenic pathway observed in metastatic and therapeutically resistant cancers, such as triple-negative breast cancer (TNBC). In preclinical models, cSNX1.3 treatment has been shown to inhibit tumor cell survival, migration, and proliferation, and to promote tumor regression and immune cell recruitment (specifically Natural Killer cells) without significant systemic toxicity.
