CSPG4 CAR-T cells are an investigational third-generation chimeric antigen receptor (CAR) T-cell therapy designed to target Chondroitin sulfate proteoglycan 4 (CSPG4), a surface antigen highly expressed in various aggressive malignancies, including chordoma, melanoma, and glioblastoma. The specific construct described in recent research utilizes a third-generation lentiviral vector incorporating CD28, OX40, and CD3ζ signaling domains to enhance T-cell persistence and potency. In the context of chordoma, these CAR-T cells are being evaluated in combination with localized hyaluronidase (PH20) delivery to degrade the hyaluronan-rich extracellular matrix, which typically acts as a physical barrier to immune cell infiltration. This combinatorial approach aims to reshape the tumor microenvironment, allowing for deeper CAR-T cell penetration and improved cytotoxic efficacy against dense solid tumors.
