CSPG4-CAR.CIK is an investigational adoptive cell therapy comprising *cytokine-induced killer lymphocytes (CIK)* genetically engineered to express a second-generation *chimeric antigen receptor (CAR)* targeting **chondroitin sulfate proteoglycan 4 (CSPG4)**[1][2][3][6]. The CAR construct incorporates **4-1BB** costimulation for enhanced activity[1][2][3]. CSPG4 is highly expressed across multiple **soft tissue sarcoma (STS)** histotypes, and CSPG4-CAR.CIK cells demonstrate potent cytotoxicity, recruitment, infiltration, and matrix penetration against STS in both two- and three-dimensional cultures and in vivo xenograft mouse models[1][2][3][6]. Primary indication: **Unresectable soft tissue sarcomas** (STS); strong preclinical evidence supports further clinical development[1][2][3][6].
