CSPG4-targeted CAR-NK cells represent an immunotherapeutic approach utilizing Natural Killer (NK) cells engineered with Chimeric Antigen Receptors (CARs). These CARs are designed to specifically recognize and bind to Chondroitin Sulfate Proteoglycan 4 (CSPG4), a transmembrane protein overexpressed on the surface of various cancer cells, including those found in melanoma and glioblastoma, while having restricted expression in normal tissues. Upon binding to CSPG4-positive cancer cells, the engineered CAR-NK cells are activated to induce targeted cytotoxicity, leading to the lysis and elimination of tumor cells. This strategy aims to leverage the inherent anti-tumor properties of NK cells, combined with the precise targeting capabilities of CAR technology, to overcome limitations of traditional therapies and reduce off-target toxicities. Preclinical studies have demonstrated the potent anti-tumor activity of CSPG4-targeted CAR-NK cells in models of glioblastoma.
