Overview

CST-3056 (also known as CuraAX) is a proprietary, selective, central nervous system (CNS)-penetrant small molecule that acts as an alpha-1A adrenoceptor (α1A-AR) partial agonist. It is being developed by CuraSen Therapeutics for the treatment of neurodegenerative diseases, with primary focus on Alzheimer's disease and neurogenic orthostatic hypotension (nOH). The drug aims to restore adrenergic signaling in the brain to improve cognitive function and address symptoms such as attention, arousal, memory, executive function, and mood. By directly activating α1A-adrenoceptors in the CNS—receptors implicated in maintaining cognitive health—CST-3056 represents a novel approach distinct from traditional therapies targeting amyloid or tau pathology. As of early 2025, CST-3056 has entered Phase 1 clinical trials to assess safety and pharmacokinetics in healthy volunteers and patients with relevant neurological conditions[1][2][3][4][5].
