Overview

CSTI-100 is a selective melanin-concentrating hormone receptor 1 (MCHR1) antagonist developed by ConSynance Therapeutics. It has completed Phase 1 clinical studies and was investigated as a potential treatment for nonalcoholic steatohepatitis (NASH), obesity, and related metabolic disorders. The drug works by blocking the MCH/MCHR1 system, which is implicated in appetite regulation, hepatic steatosis, obesity, insulin resistance, and inflammatory processes. In preclinical models of NASH with metabolic syndrome features, CSTI-100 reduced liver fat content and cholesterol levels; decreased key liver inflammation and fibrosis biomarkers; improved liver histology; promoted fat-selective weight loss through targeted reduction in food intake; and enhanced glucose tolerance and insulin sensitivity. In Phase I studies involving healthy overweight subjects, CSTI-100 was found to be safe and well-tolerated while inducing up to a 47% reduction in hunger. It also demonstrated anxiolytic effects in rodent models[3][5].
