Overview

CT-001 is an engineered recombinant Factor VIIa (FVIIa) variant protein therapeutic being developed by Coagulant Therapeutics for the treatment of acute bleeding conditions, including severe hemorrhage, postpartum hemorrhage, and trauma-induced bleeding. The molecule features specific amino acid substitutions (P10Q and K32E) in its gamma-carboxylated domain to enhance phospholipid binding and procoagulant potency. To improve safety, CT-001 is designed for rapid systemic clearance via the hepatic asialoglycoprotein receptor (ASGPR) through the incorporation of four desialylated N-glycans (T106N/N145/V253N/N322). This results in a significantly shortened half-life of approximately 5 minutes, intended to provide localized hemostatic efficacy while minimizing the risk of systemic thromboembolic events. The drug has received FDA Orphan Drug Designation for severe postpartum hemorrhage and is currently in preclinical development.
