Overview

CT-005404 is a selective dopamine transporter (DAT) inhibitor developed by Concert Pharmaceuticals. The compound was designed using Concert's proprietary deuterium chemistry platform, which involves the strategic replacement of hydrogen atoms with deuterium to enhance the metabolic stability and pharmacokinetic profile of the molecule. CT-005404 has been investigated in preclinical and early-stage research for the treatment of neuropsychiatric and neurodevelopmental disorders, including attention deficit hyperactivity disorder (ADHD) and binge eating disorder (BED). In these contexts, it acts by blocking the reuptake of dopamine, thereby increasing synaptic dopamine concentrations. While it is frequently cited in scientific literature as a reference compound for selective DAT inhibition, its current development status is unclear following the acquisition of Concert Pharmaceuticals by Sun Pharma in 2023. It is notably used as a benchmark in the development of other DAT inhibitors, such as TheraCryf's TC-114.
