**CT-03P** is a prodrug developed by Captor Therapeutics that acts as a targeted protein degrader specifically for **MCL-1 (myeloid cell leukemia 1)**, an anti-apoptotic protein overexpressed in various hematologic and solid tumors. It employs a hit-and-run degradation mechanism, rapidly inducing ~70% degradation of MCL-1 to irreversibly trigger apoptosis in cancer cells while featuring fast clearance at therapeutic doses, which differentiates it from traditional MCL-1 inhibitors by avoiding MCL-1 protein accumulation and associated cardiotoxicity. Preclinical data demonstrate superior potency compared to Novartis' MIK665 in 30 patient-derived cell lines, including those refractory to gilteritinib and venetoclax, with strong efficacy in AML models (e.g., MV-4-11 leukemia xenografts), tumor regression in combination with venetoclax, and no observed troponin-I elevations or cardiotoxicity in non-human primates even at high doses.[1][3][10][12]
