Overview

CT-05 is a small-molecule bi-functional degrader (BID) developed by Captor Therapeutics that targets Protein Kinase C theta (PKCθ). PKCθ is a serine/threonine kinase primarily expressed in T cells, NK cells, and skeletal muscle, playing a critical role in T-cell activation and proliferation. The drug functions by recruiting an E3 ligase to PKCθ, leading to its ubiquitination and subsequent proteasomal degradation. Notably, CT-05 selectively degrades the active form of PKCθ found in disease-driving cells, offering potential therapeutic benefits in autoimmune diseases (such as inflammatory bowel disease, psoriasis, and rheumatoid arthritis), oncology, transplantation, and metabolic disorders. As of 2023, the program was in pre-clinical development with plans to reach in-vivo proof of efficacy.
