Overview

CT-0525 is a first-in-class, ex vivo gene-modified autologous chimeric antigen receptor-monocyte (CAR-Monocyte) cellular therapy developed for the treatment of solid tumors that overexpress human epidermal growth factor receptor 2 (HER2). Unlike traditional CAR-macrophage therapies, CT-0525 uses monocytes that are engineered ex vivo to express an anti-HER2 CAR and are then administered to patients. These CAR-monocytes differentiate into pro-inflammatory CAR-macrophages in vivo, enabling multi-modal anti-tumor activity including improved tumor infiltration and overcoming immunosuppression within the tumor microenvironment. The therapy aims to address challenges associated with cell therapies for solid tumors such as antigen heterogeneity and limited cell persistence. Preclinical studies have shown robust reduction in tumor burden in HER2-overexpressing models. As of mid-2024, CT-0525 is being evaluated in a phase 1 clinical trial for patients with locally advanced or metastatic HER2-overexpressing solid tumors who have progressed on standard therapies[1][4][6].
