CT-1500 is an orally active small molecule drug developed by Circadian Therapeutics for the treatment of circadian rhythm sleep disorders, including Non-24 Sleep-Wake Disorder. It acts as a dual antagonist of adenosine A2A and A1 receptors (purinergic P1 receptor modulators), modulating adenosine signaling in the brain's suprachiasmatic nucleus (SCN) to influence circadian entrainment. Preclinical studies have shown that CT-1500 can lengthen the circadian period, enhance re-entrainment to shifted light-dark cycles, and phase shift circadian rhythms in animal models. The compound is clinically safe, well tolerated, orally bioavailable, and brain permeant. As of late 2024, its highest development phase is Phase 1 clinical trials in healthy volunteers for circadian rhythm sleep disorders[1][2][3][5].
