Overview

CT-202 is a preclinical-stage bispecific antibody designed to engage T cells for the treatment of solid tumors. It targets both Nectin-4, a cell adhesion molecule highly expressed in several cancers, and CD3 on T cells. The molecule incorporates two Nectin-4 binding arms and two CD3-binding single-chain Fv domains in an IgG format with a silenced Fc region, making it functionally monovalent to reduce aberrant T-cell activation and improve safety. CT-202 uses pH-dependent binding to minimize off-tumor effects by favoring tumor microenvironment conditions, aiming for high affinity and selectivity toward Nectin-4-positive cancer cells while limiting activity in normal tissues. Preclinical studies have shown potent cytotoxicity against Nectin-4-positive tumor cells with reduced cytokine release compared to benchmark antibodies. Originally developed as BA3362 by BioAtla, the asset was licensed by Context Therapeutics for further development[1][3][8].
